The present experiment was performed to study autoradiographically the effects of cycloheximide on protein synthesis of pancreas.
60 male mice, each weighed about 20gm were used, 24 mice of which were used for preliminary experiment and the rest 36 mice for main experiment. The animals were divided into experimental group and control group, then 0.2mg of cycloheximide per 100g of body weight was injected intraperitoneally to the former group. The animals were sacrificed at intervals of the 1st, 3rd, 5th, 10th, 16th and 24th days after injection. 5uc of leucine-C14 per gram of body weight was injected intraperitoneally 1 hour before sacrif ice.
The results obtained are summarized as follows:
1. The proper concentration of cycloheximide for the main experiment was 0.2mg per 100g of body weight.
2. The effects of cycloheximide on body weight were observed to decrease body weight
continuously until the 5th day and began to recover thereafter. The recovery
reached the numerical value of the first day of the experiment on the 16th day.
3. The effects of cycloheximide on the weight of pancreas were appeared with the.
increase of weight until the: 5th day. The recovery was proceeded thereafter. and
reached the same. leuel on the .24th -day with that ,of the first day of the experiment.
4. The effects of cycloheximide with leucine-C" on protein synthesis of pancreas were observed to decrease the number of silver grains from the Ist day and showed the lowest numerical value on the 5th day. The r covery was proceeded thereafter and the number of silver grains began to increas:-.
